The aim of the present work was to develop an "in vitro" sys tem capable of evaluating, at biological level, the efficacy of antineoplasic drugs that independ of previous metabolization for their action and are employed as part of treatment at the Instituto Nacional de Câncer. The assay was based on inhibition of cellular proliferation (cultured cell une K562 and PHA-stimulated lymphocytes), measured using [3H] thymidine uptake into cellular DNA. This kind of assay proved not to be adequate to test the action of Methotrexate but could be extremely sensitive when used with Vinca derivatives such as Vincristine.
